State-run media in Tehran reported that Iran had hit an unmanned U.S. vessel trying to enter an area of the Strait of Hormuz that it says is restricted.
Capt. Tim Hawkins, U.S. Central Command spokesman, told the Associated Press that the claim was a "total lie."
It is the latest salvo in a new flare-up of fighting between the two countries. The purported attack came a day after the U.S. military said forces struck three Iranian oil tankers in response to a missile attack on Navy warships.
Attacks resumed last week after roughly a month of quiet.
CENTCOM said on Sunday that U.S. forces have redirected 92 commercial vessels, disabled three and boarded two others to "ensure strict compliance" with the maritime blockade against Iran.
